Forseveral viruses a dependence on cholesterol for virus entry and/ormorphogenesis has been shown. Using flow cytometry and fluorescencemicroscopy we demonstrate that Canine distemper virus (CDV)infection of cells was not impaired after cellular cholesterol hadbeen depleted by the drug methyl-beta-cyclodextrin (MbCD). Thiseffect was independent of the multiplicity of infection (MOI) andthe cellular receptor used for infection. However, cholesteroldepletion of the viral envelope significantly reduced CDVinfectivity. Replenishment by addition of exogenous cholesterolrestored infectivity up to 80 percent. Thus, we conclude that CDVentry is dependent on cholesterol in the viral envelope.Furthermore, a reduced syncytium formation was observed when thecells were cholesterol depleted during the course of the infection.This may be related to the observation that the CDV envelopeproteins H and F partitioned into cellular detergent resistantmembranes (DRMs).
